Antibody Target Type: Pan-specific

Protein UniProt: Q9Y4A5

Protein SigNET: Q9Y4A5

Antibody Type: Polyclonal

Antibody Host Species: Rabbit

Antibody Immunogen Source: Human TRRAP sequence peptide Cat. No.: PE-01ATL99

Antibody Immunogen Sequence: CARRLYIRGHNGKIYP

Antibody Immunogen Description: Corresponds to amino acid residues A3513 to P3527

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Antibody Concentration: 0.75 mg/ml

Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol

Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Product Use: Western blotting | Antibody microarray

Antibody Dilution Recommended: 2 µg/ml for immunoblotting

Antibody Species Reactivity: Human

Antibody Positive Control: The observed molecular mass of the processed target protein on SDS-PAGE gels is reported to be around 350-450 kDa.

Scientific Background: TRRAP is a protein-serine/threonine kinase and transcription, coactivator/corepressor of the Atypical group and PIKK family. It is a multidomain kinase that belongs to the phosphoinositide-3-kinase-related kinase (PIKK) family. The TRRAP protein functions as an adaptor protein in various multiprotein chromatin complexes that collectively display histone acetyltransferase activity (HAT) and function in the epigenetic regulation of transcriptional activity. in addition, TRRAP is a component of the NuA4 histone acetyltransferase complex, which mediates the acetylation of histones H4 and H2A. TRRAP may be an oncoprotein (OP). TRRAP mediates the transcriptional activation induced by the adenovirus E1A. In addition, TRRAP plays a critical role in the regulation of Myc activation and is involved in Myc-mediated cell transformation. Several mutations (e.g. S722P in cutaneous malignant melanoma cells) in the TRRAP gene have been observed in human cancers. Knockdown of the mutant S722P TRRAP protein with siRNA in melanoma cancer cells results in increased apoptosis, indicating that the mutant TRRAP protein is essential for melanoma cell survival and tumour progression. In animal models, Cre-lox mediated knockdown of TRRAP expression lead to reduced cell proliferation due to the necessity of TRRAP for the regulation of the mitotic checkpoint and normal progression through the cell cycle.
