Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: SRPK2 (SFRSK2) is one of several protein kinases that can phosphorylate KinSubaRRGSF. It is a protein-serine/threonine kinase of the CMGC group and SRPK family. This kinase is moderate to highly expressed in most tested human tissues. It is a member of the serine/arginine (SR) protein-specific kinase family that are cell cycle-regulated protein kinases which phosphorylate SR domain-containing proteins in nuclear speckles and mediate the pre-mRNA splicing events. It phosphorylates SFRS1 and SFRS2 on serine residues. It is involved in spliceosome assembly and splicing factor trafficking. SRPK2 is activated by phosphorylation at S52 and S588. SRPK2 knock down results in hypophosphorylation of the serine/arginine domain-containing human PRP28 protein thereby destabilizing PRP28 association with the tri-snRNP. RNAi-mediated depletion in HeLa cells showed that SRPK2 is essential for cell viability, and it is required for spliceosomal B complex formation. SRPK2 has been linked with the development of glioblastoma multiforme (GM).
