Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ZC2 (TNIK; TRAF2 and NCK-interacting protein kinase) is a protein-serine/threonine kinase of the STE group and STE20 family. It may be involved in cytoskeleton regulation. Mutations at the 54, 152-153 and 171-172 positions of ZC2 lead to loss of autophosphorylation. It acts as an essential activator of the Wnt signaling pathway, and is recruited to promoters of Wnt target genes and required to activate their expression. It may act by phosphorylating TCF4/TCF7L2, and appears to function upstream of the JUN N-terminal pathway. It may mediate part of the response to environmental stress. It is part of a signaling complex composed of NEDD4, RAP2A and TNIK, which regulates neuronal dendrite extension and arborization during development.
