Scientific Background: Hsc70 (HSP73) is a member of the HSP70 family that acts as a repressor of transcriptional activation. Isoform 2 of Hsc70 may function as an endogenous inhibitory regulator of Hsc70 by competing with co-chaperones. It interacts with BAG1, DNAJC7, HERC5, HSPH1/HSP105, IRAK1BP1, PACRG, SV40 VP1, and TSC2, and can be found in a mRNP granule complex. Hsc70 also binds CITED1 and this interaction suppresses the association of CITED1 to p300/CBP and Smad-mediated transcription transactivation. Upon DNA damage, Hsc70 undergoes increased phosphorylation, apparently by ATM or ATR.
