Scientific Background: ErbB2 is a receptor-tyrosine kinase of the EGFR family, which apparently needs a coreceptor for ligand binding. It is an essential component of a neuregulin-receptor complex, although neuregulins do not interact with it alone. It is not directly activated by EGF, TGF- alpha and amphiregulin, although GP30 is a potential ligand for this receptor. Upon ErbB2 activation, the MEMO1-RHOA-DIAPH1 signalling pathway elicits the phosphorylation and inhibition of GSK3B at cell membrane. This prevents the phosphorylation of CLASP2 and APC, allowing its association with the cell membrane. In turn, membrane-bound APC allows the localization of MACF1 to the cell membrane, which is required for microtubule capture and stabilization. The tyrosine-phosphorylated ErbB2 receptor also interacts with PIK3C2B, PRKCABP, MUC1, GRB7, ERBB2IP, Src, KPNB1, RANBP2, EEA1, CRM1, CLTC, PTK6, RPA94 and ACTB. ErbB2 is amplified in ~30% of breast cancer and the overexpression induces constitutive activity, which correlates with poor survival. The antibody Herceptin is approved for treatment of metastatic breast cancer with HER2 amplification/overexpression. Somatic mutations seen in 4% of lung cancers and also in breast, gastric, ovarian cancer and glioblastoma. Inhibitors of ErbB2 include Herceptin, lapatinib, PKI-166, EKB-569, and CI-1033. Four alternatively spliced human isoforms of ErbB2 have molecular masses of 137,910, 136,501, 70,917 and 62,568 Da.
