Scientific Background: IGF1R is one of several protein kinases that can phosphorylate IRS1 (891-902). Human IGF1R is a receptor protein-tyrosine kinase of 1367 amino acid length, with a predicted molecular mass of 154,793 Da. It is a member of the TK group of protein kinases in the InsR family. InsR is post-translationally cleaved into two chains, alpha and beta that are covalently linked. This kinase is highly expressed and widely distributed in most tested human tissues. Orthologues of IGFR1 are commonly conserved in vertebrates. IGF1R is activated by binding insulin-like growth factor I (IGF1), which induces the pre-dimerization receptor to autophosphorylation. Phosphorylation of Tyr-1161 and Tyr-1346 increases its phosphotransferase activity and induces interaction with Src. Phosphorylation of Tyr-980, Tyr-1161, Tyr-1165 and Tyr-1166 increases its phosphotransferase activity.
