Scientific Background: Obscn is a protein-serine/threonine kinase of the CAMK group and Trio family. It is a protein kinase, and a cytoskeletal calmodulin and titin-interacting RhoGEF. It is part of a family of giant sacromeric signalling proteins that includes titin and nebulin, and has been implicated in the organization of myofibrils during assembly and mediation of interactions between the the sarcoplasmic reticulum and myofibrils. It seems to be an adapter protein that is involved in assembly of myosin into sarcomeric A bands in striated muscle. Isoform 3 together with ANK1 isoform Mu17/Ank1.5 may provide a molecular link between the sarcoplasmic reticulum and myofibrils. Although it demonstrates a high rate of mutation in cancer cells, it is unclear how this implicates in oncogenesis. A chromosomal aberration involving Obscn has been reported in Wilms tumour, and the gene has a translocation t(1;7)(q42;p15) with PTHB1. Due to its large size, there is a greater chance of mutations being detected in this protein in human cancers, and these may be mainly bystander mutations.
