Scientific Background: IRR is a receptor protein-tyrosine kinase that is a member of the TK group of protein kinases in the InsR family. IRR is required for the appearance of male gonads and for male sexual differentiation. It can sense pH in the extracellular environment. When IRR is activated a signalling cascade is started which involves IRS1 and PKB/Akt1. A mutation that can inhibit IRR autophosphorylation is Y1145F with Y1146F. Expression of IRR mRNA is restricted to a few tissue types and has been detected in neuronal tissues as well as in neuroblastomas. As a result of its occurrence in neuroblastomas, it has speculated that IRR may be involved in tumorigenesis. RT-PCR and solution hybridization/RNAse protection assay have shown strong coupling of IRR mRNA in 18 tumour cell lines. IRR has also been linked with the development of glioblastoma multiforme and lung adenocarcinomas.
