Scientific Background: MRCK-a (CDC42BPA, CDC42-binding protein kinase A) is a protein-serine/threonine kinase of the AGC group and DMPK family. It is a critical downstream effector and plays a role in the regulation of cytoskeleton remodelling and cell migration. Agonist binding to the phorbol ester binding site disrupts this, releasing the kinase domain to allow N-terminus-mediated dimerization and kinase activation by transautophosphorylation. MRCK-a is maintained in an inactive, closed conformation by an interaction between the kinase domain and the negative autoregulatory C-terminal coiled-coil region.
