Scientific Background: PFTAIRE1 (CDK14, PFTK1) is a protein-serine/threonine kinase of the CMGC group and CDK family. It is expressed primarily in the postnatal and adult nervous system, and regulates cycle progression and cell proliferation. It is expressed at high levels in brain, pancreas, kidney, heart, testis, and ovary. Its phosphotransferase activity is stimulated by association with cyclins CCDN3 and CCNY. It binds to 14-3-3 isoforms when it is phosphorylated at S119, which can be enhanced by CCNY. It is repressed by CDKN1A. Insertional mutagenesis studies in mice support a role for this protein kinase in mouse cancer oncogenesis. Positive expression of PFTAIRE1 when compared to its negative expression predicts resistance to chemotherapy and poorer prognosis in patients with oesophageal squamous cell carcinomas over a 5-year period.
