Scientific Background: MAPKAPK3 is one of several protein kinases that can phosphorylate KinSub1RRDSF. Human MAPKAPK3 is a protein-serine/threonine kinase of 382 amino acid length, with a predicted molecular mass of 42,987 Da. It is a member of the CAMK group of protein kinases in the MAPKAPK family, and MAPKAPK subfamily. This kinase is highly expressed and widely distributed in most tested human tissues. Orthologues of MAPKAPK3 are highly conserved in vertebrates and insects. MAPKAPK3 interacts with TCF3 and with polycomb proteins, such as PCH2 and BMI1/PCGF4. MAPKAPK3 is closely related to MAPKAPK2 sharing 72% nucleotide and 75% amino acid identity (1). MAPKAPK3 is activated by growth inducers and stress stimulation of cells. In vitro studies have demonstrated that ERK1/2, p38 MAP kinase and Jun N-terminal kinase were all able to phosphorylate and activate this kinase, which indicated the role of MAPKAPK3 as an integrative element of signalling in both mitogen and stress responses (2). MAPKAPK3 has been linked with the development of glioblastoma multiforme (GM).
