Scientific Background: ROCK2 (ROK-alpha) is a protein-serine/threonine kinase of the AGC group and DMPK family. It regulates the actin cytoskeleton and cell polarity,and is involved in the regulation of smooth muscle contraction, formation of stress fibre and focal adhesions, cell adhesion, and cell motility. It is activated by binding to the small G protein RhoA, and by phosphorylation at T967 and S1099. It is inhibited by phosphorylation at Y722. Insertional mutagenesis studies in mice support a role for this protein kinase in mouse cancer oncogenesis.
