Scientific Background: MKK3 (MAP2K3, MEK3) is a Dual specificity protein kinase of the STE group and STE7 family. MKK3 is activated by cytokines and environmental stress in vivo. Phosphorylation of S218 and T222 increases its phosphotransferase activity. S218A and T222A substitutions are involved in inactivation. S218E and T222E mutations are associated in constitutive activation. It catalyzes the concomitant phosphorylation of a threonine and a tyrosine residue in the activation T loops of the p38 MAP kinase isoforms. Defects in the MKK3 gene may be involved colon cancer.
