Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.This antibody was also subject to negative purification over phosphotyrosine-agarose.

Scientific Background: TNK1 is a protein-tyrosine kinase of the TK group and Ack family. It is a non-receptor kinase that functions as a negative regulator of cell growth through inhibition of the Ras-MAPK intracellular signalling pathway. TNK1 is thought to function in many tissues during fetal development and more selectively in the adult, with activity limited to cells of the lymphohematopoietic system. In addition, TNK1 is also predicted to play a role in phospholipid signalling. Phosphorylation of the TNK1 protein at several sites (Y77, Y235, Y277, Y287, and Y353) was observed in Hodgkin lymphoma (HL) cancer cell lines, indicating a possible role for the aberrant activity of the protein in the pathogenesis of this disease. In addition, a truncated form of TNK1 was identified in HL cancer cells. However, the effect of the truncation on its phosphotransferase activity was unclear.
