Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.This antibody was also subject to negative purification over phosphotyrosine-agarose.

Scientific Background: TXK is one of several protein kinases that can phosphorylate KinSub1MTMTP. TXK (RLK) is a protein-tyrosine kinase that is a member of the TK group of protein kinases in the Tec family. This kinase is moderate to highly expressed in most tested human tissues except in the brain, heart and spinal cord where it is poorly expressed. TXK has been shown to participate in multiple intracellular signalling pathways and thus influences many cellular processes, including the regulation of the actin cytoskeleton. TXK functions to regulate the development, activity, and differentiation of T-cells and NKT-cells. It is an important component of signalling pathways downstream of lymphocyte antigen receptor. TXK is phosphorylated in response to T-cell receptor stimulation and can be activated by phosphorylation by Src family kinases. The TXK protein is activated by phosphorylation at Y420. . Excessive TXK protein expression is seen in patients with Behcet's disease. Over production of TXK leads to increased Th1 cell function that is involved in the pathogenesis of Behcet's disease.
