Peptide Name: TXK (417-423) pY420
Product Use: Services as a blocking peptide for use with the TXK-pY420 rabbit polyclonal antibody (Cat. No.: AB-PK844) that is also available from Kinexus. This phosphopeptide may also be useful as a substrate for screening the phosphatase activity of protein phosphatases. Y420 phosphorylation is predicted to be stimulatory for phosphotransferase activity. This phosphosite is located in the kinase activation loop between catalytic subdomains VII and VIII.
Peptide Production Method: Solid-phase peptide synthesis
Peptide Origin: Homo sapiens
Peptide Sequence: DDE-pY-VSS
Peptide Modifications N Terminus: Free amino
Peptide Modifications C Terminus: βAla-Cys
Scientific Background: TXK is a non-receptor protein-tyrosine kinase which largely overlaps in function with ITK in the regulation of the adaptive immune response. TXK functions to regulate the development, activity, and differentiation of T-cells and NKT-cells. The TXK protein is activated by phosphorylation of the protein at the Tyr-420 residue, which results from a series of events triggered by the activation of T-cell receptors (TCR) by antigen presenting cells (APC). TXK has been shown to participate in multiple intracellular signalling pathways and thus influences many cellular processes, specifically the regulation of the actin cytoskeleton.
