Scientific Background: Lck is a non-receptor protein-tyrosine kinase of the TK group and Src family. Phosphorylation of Y394 increases phosphotransferase activity and induces interaction with Lck. Phosphorylation of Y192 and Y505 inhibits Lck phosphotransferase activity. Lck has a critical function in the selection and maturation of developing T-cells in the thymus, as well as in the function of mature T-cells throughout the body. Lck is a key signalling component of the T-cell antigen receptor (TCR)-linked signal transduction pathway and is constitutively bound to the cytoplasmic domains of the CD4 and CD8 membrane receptors. Lck is expressed through all stages of thymocyte development and is necessary for regulating both pre-TCR and mature alpha beta TCR-mediated maturation. Lck may be an oncoprotein (OP) based on its similarity to other Src family protein-tyrosine kinases.
